TOPRAK VE RUMENDEN İZOLE EDİLEN BAZI CLOSTRIDIUM SP. İZOLATLARINDA SELÜLAZ ETKİNLİKLERİNİN BELİRLENMESİ

Bu çalışmada, toprak ve rumenden izole edilen Clostridium suşlarının selülaz etkinlikleri araştırılmıştır. En yüksek selülaz etkinliği gösteren Clostridium sp. CF147 suşundan selülaz izolasyonu yapılarak SDS-PAGE profilleri saptanmıştır. Selülazın sırasıyla 81000 Da ve 66000 Da moleküler ağırlığa sahip iki banttan, oluştuğu tespit edilmiştir. Bu enzimin pH aralığı ve sıcaklık (termal) kararlılığı belirlenmiştir. Selülazın en uygun pH’sı 7.0 olarak bulunmuştur. Selülazın 55?C’de %64 oranında kararlı olduğu tespit edilmiştir.

DETERMINATION OF CELLULASE ACTIVITIES IN SOME CLOSTRIDIUM SP. ISOLATES WHICH ISOLATED FROM SOIL AND RUMEN

In this study, it was investigated for cellulase activities of Clostridium strains isolated from soil and rumen. Cellulase isolation was performed and SDS-PAGE profiles were determined from Clostridium CF147 strain which showed the highest cellulase activity. It was determined that cellulase consists of two bands with molecular weight of 81000 Da and 66000 Da, respectively. pH range and thermal stability of this enzyme were determined. Optimum pH value of cellulase was found as 7.0. Cellulase was determined the stable at 55?C upto 64%.
