ÇEŞİTLİ TANI GRUPLARINDA ANTİ-NÖTROFİL SİTOPLAZMİK ANTİKOR POZİTİF PATENLERİN DEĞERLENDİRİLMESİ

Bu çalışmada, çeşitli tanılarla gönderilen örneklerde antinötrofil sitoplazmik antikorların (ANCA) sıklığı, paternleri ve klinik durumlarla ilişkisi araştırıldı. Toplam 1668 kan örneğinde ANCA varlığı retrospektif olarak araştırıldı. Tüm serumlar önce indirekt immunofloresan (IIF) tekniği ile (Euroimmun, Lübeck, Almanya) çalışıldı. Pozitif floresan veren örnekler spesifik antijenleri belirlemek için öncelikle en sık saptanan antijenler olan proteinaz–3 (PR3) ve myeloperoksidaz (MPO) antijenlerini içeren enzyme-linked immunosorbent assay (ELISA) ile (Euroimmun, Lübeck, Almanya) test edildi. Bu antijenlerden herhangi biri saptanamayan serum örnekleri daha nadir görülen antijenler olan katepsin G, laktoferrin, elastaz ve bakterisidal permeabilite arttırıcı proteini (BPI) içeren ELISA ANCA profil testi ile (Euroimmun, Lübeck, Almanya) tekrar çalışıldı. Tüm serum örneklerinin 320’sinde (%19.2) IIF tekniği ile pozitif boyanma saptandı. ELISA testi ile 49 hastada spesifik antijen belirlen

ASSESMENT OF ANTI-NEUTROPHIL CYTOPLASMIC ANTIBODY POSITIVITY PATTERN IN VARIOUS DIAGNOSTIC GROUPS

In present study, frequency and patterns of anti-neutrophil cytoplasmic antibody (ANCA) and its` relationship with clinic situations were investigated in samples which were referred by miscellaneous diagnosis. Presence of ANCA were retrospectively studied on 1668 samples. All sera were first evaluated by indirect immunofluorescence technique (IIF). Then, samples with positive florescence were evaluated to determine specific antigens by enzyme-linked immunosorbent assay (ELISA) which implies proteinase–3 and myeloperoxidase. Samples in which none of these antigens were detected were reevaluated by ELISA ANCA profile test which implies less common antigens such as cathepsin G, lactoferrin, elastase and bactericidal permeability-increasing protein. Positive staining was found in 320 (19.2%) of 1668 serum samples by IIF technique. Samples in which specific antigens were found by ELISA were obtained from 49 patients.
